Abstract
Precision medicine is critically dependent on better methods of diagnosing and stagingdisease and predicting drug response. Histopathology using hematoxylin and eosin (H&E)-stained tissue remains the primary diagnostic method for cancer. Recently developed highlymultiplexed tissue imaging methods promise to improve research studies and clinical practicewith accurate, spatially resolved single-cell data. One of these is RareCyte's Orion platform, which enables the collection of H&E and highly complex immunofluorescence images fromthe same cells in a whole slide format suitable for diagnostics. This approach enables rapid IFstaining of up to 18 biomarkers in a single step.
The Institute of Clinical and Molecular Pathology, Faculty of Medicine, UPOL, was selectedby RareCyte to loan the platform for 1 year. In cooperation with the staff of this company, ourtask is to make this equipment operational. To date, we have a panel of 8 antibodiesconjugated with immunofluorescent label, comprising: CD3e, CD8a, CD68, CD163, E-cadherin, FAP, FOXP3 and TIGIT. Almost all antibodies work relatively well except for theCD68 marker, for which we are still looking for a way to obtain optimal staining.
